The Cell Imaging Core Facilities' mission has been to provide state-of-the-art cell imaging technologies to SKCCC members. Services include light and fluorescence microscopy, stereo and confocal microscopy, infrared imaging, time lapse, and laser microdissection. Images can be acquired using a variety of video and CCD cameras, and then analyzed and manipulated with cutting edge image analysis software programs. Users receive technical support by the core personnel and detailed instruction in the use of all core equipment.
Brightfield and fluorescent microscopy
All of the microscopes in our facility are brightfield as well as fluorescent capable. A variety of filter cubes allows the identification and visualization distinct populations of stained cells. A choice of either a high resolution black and white ccd camera or a color camera allows the acquisition of publication quality microscopic images. Comprehensive software allows for quantitative analysis.
Odyssey ®- Infrared Imaging System
With direct infrared detection, the Odyssey System quantifies low-abundance proteins. Two detection channels allow the simultaneous detection and analysis of two separate targets on the same gel. Other applications include in-cell western and tissue imaging.

Confocal Microscopy
Our most recent acquisition is the Nikon C1si True Spectral Imaging Confocal Laser Scanning Microscope System. One of the advantages of conventional Confocal microscopy is the ability to optically section tissue or cells therefore eliminating out-of-focus glare. The C1si also has the capability of acquiring 32 channels of fluorescent spectra over a 320 nm wide wavelength in a single pass using spectral imaging.
Timelapse Microscopy
The Nikon TE2000E inverted microscope is equipped with the Live Cell Incubation system, which is very stable and allows for optimally adjusted CO2 flow. Extended time lapse data can be acquired and analyzed with Elements software.
Stereoscopic Microscopy
The Nikon SMZ 1500 fluorescent stereoscopic zoom microscope is suitable for quickly assessing the presence of fluorescent proteins, such as GFP in live cells. In addition, the low magnification capability permits photography of whole tissue mounts.
Laser Capture Microdissection
The Leica LMD 7000 has proven to be a valuable asset to the Core users. With the ability to dissect out discrete cells or cellular components as well as defined areas of tissue, this apparatus provides the means for obtaining pure populations of cell types from histological sections, thereby facilitating a broad range of projects.
Cameras and Imaging Software
The facility provides access to four black and white high resolution CCD cameras as well as a 12-million-pixel digital Nikon DXM 1200F. The different camera types are set up to be used on any of the microscopes in the facility. Advanced software packages are available for image acquisition and analysis, ACT-1 and Elements.
